    Explanation of the Test:
    The immune response, that normally protects dog from infection may be harmful for organism. Immunological reactions connected with allergies are quite complicated. Most of the reactions are related with the change of the fraction of IgE antibodies in blood. During allergic reaction proteins attach to IgE antibodies and after connection with mast cells, they cause histamine release. That has direct impact on inflammation formation. That is why the IgE level in canine blood is so important for prognose of dog’s health. our Canine total IgE rapid Test is based on one-step lateral flow technique through chromatographic strip. The solution moves on the membrane with capillary motion. During procedure solution after incubation containing IgE reacts with antibodies labeled with colloidal gold, that are immobilized on the membrane. The presence of IgE above threshold sensitivity is visible as colorful reaction on test line. In all cases also "C" line has to appear, that proves the proper procedure of the Test.

    Principle:
    Canine total IgE Rapid Test is based on sandwich lateral flow Immunochromatographic assay.
    Specimen preparation and storage:
    Specimen should be obtained and treated as below;
    - Serum or plasma: collect the whole blood from the patient dog, centrifuge it to get the serum, or place the whole blood into a tube which contains anticoagulants to get plasma.
    - All specimen should be tested immediately. If not for testing right now,
    they should be stored at 2-8℃ for maximum of 48 hours.
    Test Procedure using Serum:
    1- Allow all materials, including specimen and Test strip, recover to 15-25℃ before running the assay.
    2- draw 1cc of blood from your dog.
    3- centrifuge for 3-5 minutes at 4000rpm to separate the serum. Or standing the whole blood Tube for two hours at 4ºC to separate the serum.
    4- take out the Test strip from the foil pouch and place it horizontally.
    5- using a dropper add 1 drop of sample into diluent buffer Tube.
    6- recap the assay diluent buffer tube and mix thoroughly by inverting the buffer Tube for a few seconds.
    7- using a disposable dropper, add 3 drops of mixed sample into sample hole (“S”), drop by drop vertically.
    8- interpret test results at 5-10 minutes. Do not interpret after 10 minutes.
    Test procedure using Fecal (stool) sample:
    1-
    Coat the sampling swab with a thin layer of fecal material or saturate the swab if the sample is liquid. (
    The provided sampling swab is not intended for rectal use)
    2- insert the swab stick into the assay diluent Tube.
    3- recap the assay diluent Tube and mix thoroughly by inverting the buffer Tube for a few seconds.
    4- take out the Test strip from the foil pouch and place it horizontally.
    5- using the provided dropper gradually drip 2-3 drop of the sample extraction into the Test well.
    6- interpret the result in 5-10 minutes, result after 10 minutes is considered as invalid.
    Interpretation of Results:
    - Positive (+)
    : The presence of both “C” line and zone “T” line, no matter T line is clear or vague. The result indicates that the total IgE
    is above 10μg/mL (it is recommended that the Veterinarian performs a
    specific IgE Test)
    - Negative (-):
    Only clear C line appear. No T line observed. The
    result indicates that the total IgE is below 10μg/mL.
    - Invalid:
    No colored line appears in C zone. No matter if T line
    appears.
    Storage and stability:
    - The kit can be stored at room temperature (4-30°C).
    - The test kit is stable
    - DO NOT FREEZE and Do not store the test kit in direct sunlight.
